Anti-Platet Aggregation Test in Vitro of Tembelekan (Lantana Camara L.), Urang Aring (Eclipta Prostrata (L.) L.) and Kenikir (Cosmos Causatus Kunth.) Leaves Ectract
Objective: The leaf of tembelekan, urang aring and kenikir have flavonoid, saponin and atsiri oil. These compounds have a role in inhibiting the process of platelet aggregation. At this research, tembelekan, urang aring and kenikir leaves extracted with maceration methode using ethanol 70% as a solvent. This study aims to determine the potential of antiplatelet aggregation from that three extracts and compare which extracts have the largest percentage of inhibition.
Methods: The samples consist clopidogrel 2 mg/ml as positive contol, negative control of the aquadest and extract test with concentrations 40, 60, 80 and 100 mg/ml with additions of Platelet Rich Plasma (PRP) for each concentration. The absorbance test sample is measured at 600 nm, before the ADP adds 5 mM to 20 μL. After ADP added, then measured again the plasmaabsorbance of the same wavelength. Antiplatelet aggregation test was performed by plasma absorbance calculating before and after adding Adenosine diphosphate (ADP). Results: The test show percentage inhibition of platelet aggregation concentrations 100, 80, 60 and 40 mg/ml respectively are tembelekan leaf extract (91,17%; 73,27%; 72,86% and 37,15%), urang aring leaf extract (74,55%; 65,60%; 48,00% dan 39,08%) and kenikir leaf extract (59,17%; 52,56%; 14,25% dan 13,82%). There were no significant difference between tembelekan of concentration 100 mg/ml with positive control (clopidogrel) (p=0,513). Conclusion: These three extracts have potential to inhibit platelet aggregation and the biggest potential for inhibiting platelet aggregation is tembelekan leaf extract.
