Validation of Simultaneous Identification Methods of Ibuprophen, Mefenamic Acid, Phenylbutazone, Dexamethasone, and Prednisolone in Herbal Medicine Intended for Aches and Pain by High Performance Liquid Chromatography



Jamu has been used for generations by the Indonesian people for certain health
purposes. Based on the results of the supervision of the Indonesian National
Agency of Drug and Food Control, there are still many herbs containing
adulterants circulating in the community. The purpose of this study was to
obtain a valid method for the simultaneous identification of ibuprofen,
mefenamic acid, phenylbutazone, dexamethasone, and prednisolone in jamu
intended for aches and pain using reverse phase high performance liquid
chromatography method using C18 column stationary phase with a column
length of 250 mm, inner diameter of 4,6 mm, 5 µm particle size, acetonitrileammonium
acetate

nm wavelength. Method validation included specificity test and
determination of detection limit. The results of this study indicated that this
method has detection limit of prednisolone, dexamethasone, phenylbutazone,
mefenamic acid, and ibuprofen of 0,91; 0,92; 0,92; 1,62; and 10,67 µg/mL,
respectively. Solid phase extraction had been used as sample preparation with
the average recovery results of prednisolone, dexamethasone, phenylbutazone,
mefenamic acid, and ibuprofen were 98,63; 109,39; 98,45; 63,99; 89,82%,
respectively. This HPLC identification method was valid for the identification
of prednisolone, dexamethasone, phenylbutazone, mefenamic acid, and
ibuprofen in jamu but the SPE sample preparation method required further
research to obtain the optimum mefenamic acid extraction method.
